چکیده

The yeast Saccharomyces cerevisiae used in brewing and bread making is a unicellular microorganism and serves as a model system of the eukaryotic cell. Genetic and molecular biochemical studies also provided pioneering discoveries in the field of vesicular trafficking. Recently, our understanding of this field has greatly advanced, however, there still remain unsolved problems. For example, since vesicular trafficking of each component occurs between related organelles, how the Golgi compartments keep their identity as a distinct organelle remains to be solved. Therefore, we carried out studies to investigate these problems.

Localization of components involved in protein transport and processing through the yeast Golgi apparatus

Saccharomyces cerevisiae sec7 mutants exhibit pleiotropic deficiencies in the transit of proteins through the Golgi apparatus, and elaborate an array of Golgi apparatus-like cisternae at a restrictive growth temperature (37 degrees C). The SEC7 gene encodes an essential high-molecular weight protein (227 kD) that is phosphorylated in vivo. In cell lysates, Sec7 protein (Sec7p) is recovered in b...

Characterization of YIPF3 and YIPF4, cis-Golgi Localizing Yip domain family proteins.

The Yip1 domain family (YIPF) proteins are homologues of yeast Yip1p and Yif1p, which are proposed to function in ER to Golgi transport. Here, we report the characterization of YIPF3 and YIPF4, homologues of human Yif1p and Yip1p, respectively. Immunofluorescence and immuno-electron microscopy showed that both YIPF3 and YIPF4 are clearly concentrated in the cis-Golgi. While YIPF4 was detected a...

Yeast Kex1p is a Golgi-associated membrane protein: deletions in a cytoplasmic targeting domain result in mislocalization to the vacuolar membrane

We have investigated the localization of Kex1p, a type I transmembrane carboxypeptidase involved in precursor processing within the yeast secretory pathway. Indirect immunofluorescence demonstrated the presence of Kex1p in a punctate organelle resembling the yeast Golgi apparatus as identified by Kex2p and Sec7p (Franzusoff, A., K. Redding, J. Crosby, R. S. Fuller, and R. Schekman. 1991. J. Cel...

Sac1–Vps74 structure reveals a mechanism to terminate phosphoinositide signaling in the Golgi apparatus

Sac1 is a phosphoinositide phosphatase of the endoplasmic reticulum and Golgi apparatus that controls organelle membrane composition principally via regulation of phosphatidylinositol 4-phosphate signaling. We present a characterization of the structure of the N-terminal portion of yeast Sac1, containing the conserved Sac1 homology domain, in complex with Vps74, a phosphatidylinositol 4-kinase ...

The C-TERMINUS of AtGRIP Is Crucial for Its Self-Association and for Targeting to Golgi Stacks in Arabidopsis

BACKGROUND In animals and fungi, dimerization is crucial for targeting GRIP domain proteins to the Golgi apparatus. Only one gene in the Arabidopsis genome, AtGRIP, codes for a GRIP domain protein. It remains unclear whether AtGRIP has properties similar to those of GRIP domain proteins. RESULTS In this study, western blot and yeast two-hybrid analyses indicated that AtGRIPs could form a para...
